    Abstract: 2-Methylisoborneol(2-MIB)isacommonlydetectedcyanobacterialodorantindrinkingwatersourcesin manycountries.Toprovidesafeandhigh-qualitywater,developmentofamonitoringmethodforthe detection of2-MIB-synthesis(mibC) genesisveryimportant.Inthisstudy,newprimersMIBS02F/Rin- tended specifically forthe mibC geneweredevelopedandtested.Experimentalresultsshowthatthe MIBS02F/R primersetwasabletocapture132-MIBproducingcyanobacterialstrainsgrowninthela- boratory,andtoeffectivelyamplifythetargetedDNAregionfrom172-MIB-producingcyanobacterial strains listedintheliterature.TheprimerswerefurthercoupledwithaTaqManprobetodetect2-MIB producers in29drinkingwaterreservoirs(DWRs).Theresultsshowedstatisticallysignificant correla- tions between mibC genesand2-MIBconcentrationsforthedatafromeachreservoir(R2¼0.413–0.998; po0.05), fromallreservoirsineachofthethreeislands(R2¼0.302–0.796; po0.01),andfromalldataof the threeislands(R2¼0.473–0.479; po0.01).Theresultsdemonstratethatthereal-timePCRcanbean alternativemethodtoprovideinformationtomanagersofreservoirsandwaterutilitiesfacing2-MIB- relatedincidents.
