    Title: 珊瑚純化物11-dehydrosinulariolide誘導人類膀胱癌細胞生長停滯及細胞凋亡

    Abstract: 11-dehydrosinulariolide 從珊瑚Sinularia flexibilis 分離純化的化合物,過去研究顯示能抑制口腔癌細胞Ca9-22和人類黑色素瘤細胞A20585之生長,具有抗腫瘤增殖能力。本研究計畫擬探討11-dehydrosinulariolide 對人類的膀胱癌細胞J82和BFTC-909生長之影響。目的希望瞭解11-dehydrosinulariolide 抑制膀胱癌細胞生長和誘發細胞凋亡的機轉及可能訊息傳遞路徑。實驗方法採用MTT 測定細胞存活率、TUNEL/DAPI 螢光染色法和免疫染色法分析蛋白質表現。結果顯示隨著當11-dehydrosinulariolide 濃度增加到27μM 時,開始抑制膀胱癌細胞J82和BFTC-909的生長,且在TUNEL/DAPI 染色實驗,11-dehydrosinulariolide 能誘導細胞走向凋亡。得知在BFTC-909和J82細胞的粒線體失調相關的凋亡蛋白(如Bad, Bax 和p-Bcl2)受到活化,以及內質網壓力相關蛋白,如GRP78、calnexin、p-PERK、p-eIF2α、ATF4、CHOP、ATF6均受到活化。此外,也發現11-dehydrosinulariolide 會引發PI3K-AKT-mTOR 和MAPK訊息傳遞路徑影響膀胱癌細胞的增殖、分化與轉移,結果表明它有可能被開發為治療劑人類膀胱癌的藥物。
