    Title: 可可發酵過程菌種篩選

    Abstract: 我們從發酵167小時的可可豆樣本中,針對 16S megagenomics區間進行序列定
    序,共定序出總長度分別為13.1 Mb(CC1)及53.8 Mb(CC2)。結果發現:在CC2樣品序列比對結果發現,數量前三大之OUT菌群與CC1樣品序列分析結果相同,樣品之菌相第一為Acetobacteraceae (醋桿菌)為最大宗,第二為Enterobacteriaceae (腸內細菌),第三為Paenibacillaceae (類芽孢桿菌),而三個菌科 OTU總合佔所有 OUT之 97%。屬於發酵前期之乳酸菌至發酵終點時,族群數量變少,故不易篩選。以MRS培養基在厭氧環境下,篩選出的未知菌株,透過 EMB鑑別培養基培養後,出現綠色金屬光澤,以此判定為 E. coli,乳酸菌No1~3皆為 E. coli。後期多以醋酸菌為主發酵, No1, 10, 11皆為醋酸菌,經序列比對為 Acetobacter pasteurianus (100% identities);而 No 9為G(+)菌 Bacillus thuringiensis (100% identities)。真菌選殖過程中未出現酵母菌,但出現單一綠黴菌,此菌為果腐黴菌,可能可可樹於種植過程中曾遭受真菌感染。
