    Title: 研究牛樟芝三萜類化合物25S-antcin K於類風溼性關節炎之療效

    Abstract: 類風濕性關節炎(RA, Rheumatoid arthritis)是一種廣為人知的自體免疫疾病,與關節疼痛、腫脹、軟骨與骨骼退化以及關節畸形相關。在細胞與動物模型研究中,台灣傳統藥用真菌牛樟芝(Antrodia cinnamomea)的萃取物顯示出抗發炎特性。先前研究從牛樟芝中分離出25(S)型麥角甾烷類(ergostane) 之樟芝酸K(antcin K)立體異構物,即25S-antcin K,並發現其活性較樟芝酸K更強。在本研究中,我們探討了25S-antcin K於類風溼性關節炎之療效。
    結果顯示,25S-antcin K可透過抑制AMPK、Akt及p38訊號通路並促進miR-374b表現,進而阻斷IL-1誘導的TNF-、CCL2及VEGF等關節炎相關因子的產生。此外,25S-antcin K的給予亦能有效減輕膠原蛋白誘導性關節炎(CIA)所引發的RA症狀,包括發炎反應、軟骨退化與骨侵蝕。這些結果提供了一條新的研究途徑,顯示25S-antcin K可作為發展RA治療藥物的潛在候選分子。
