    Title: 大益牛樟芝抑制人類肝癌細胞HepG2及抗人類B型肝炎病毒活性之探討

    Abstract: 本報告以體外細胞培養模式,利用酵素連結免疫吸附分析法(Enzyme-Linked Immunosorbent Assay; ELISA)探討大益牛樟芝萃取物抑制肝癌細胞HepG2及抗人類B型肝炎病毒活性。MTT assay測試結果顯示,對肝癌細胞(HepG2)給藥處理48小時,國內段木子實體IC50為39.8g/mL、國內段木菌片IC50為32.5g/mL及進口段木菌片IC50為28.5g/mL,進口段木菌片作用最強。細胞凋亡藉由ELISA kit測定,顯示三者均具有誘導肝癌細胞凋亡之活性。更進一步利用HepG2.2.15作為野生型HBV產生之病毒株,M33作為肝安能抗藥性突變株,進行抗人類B型肝炎病毒活性之探討,結果顯示,國內段木子實體與國內段木菌片,無細胞毒性下,對B型肝炎野生型病毒株(HepG2.2.15) 及肝安能抗藥性突變株(M33),表面抗原具有輕度抑制作用;進口段木菌片無細胞毒性下,對B型肝炎野生型病毒株(HepG2.2.15),表面抗原具有輕度抑制作用;對肝安能抗藥性突變株(M33) 表面抗原具有中度抑制作用;三者對e抗原均無抑制作用。
